ISSR based analysis of genetic variability of plantlets culture of pineapple (Ananas comosus L.) from Sipahutar, North Sumatera, Indonesia

Sipahutar pineapple is very popular in North Sumatra, because of the distinctive sweet taste and normal water content. Furthermore, it large size has a yellow fruit skin color with greenish tips. However, problem production limited amount good quality seeds. The utilization vitro culture techniques on pineapples allows emergence somaclonal variations, especially plantlets that have undergone subculture stage. This variation one problems commercial seedling production, therefore, necessary to conduct an initial examination using inter simple sequence repeat (ISSR) molecular marker. study aimed determine genetic primary stability ISSR, which can be used plants native Sipahutar. methods include sample preparation, DNA isolation, primer optimization, ISSR amplification by PCR method, electrophoresis. total 15 samples were amplified six primers, data analyzed cluster method NTSYS-PC software. final result was visualized dendrogram analysis diversity conducted GenAlex. results showed level variability pineapple, markers 76-97%. Meanwhile, influenced long period use N6-benzyladenine. primers observe variability, except for 25 PIC value 0.000.
